Long Read Sequencing is also referred to as third-generation sequencers, are used to directly sequence single molecules of DNA in real time. It is often carried out without the need for amplification. The method of direct sequencing allows the production of comparatively longer reads than those from Short Read Sequencing. It allows for the retrieval of longer sequence which is greater than (> 10,000 bp). Other ‘synthetic’ LRS approach uses modified sample processing and SRS to allow computationally reconstruct long reads from shorter sequencing tools. Long reads can develop de novo assembly, transcript isoform identification, detection of structural variants and mapping certainty. These competences, together with continuing improvement in throughput, accuracy, and cost reduction, have made long-read sequencing an option for a broad range of applications in genomics for both model and non-model organisms.
